چکیده

Photocycloaddition, Thymine, Azetidine, Cyclobutane Adducts Photochemical reactions of thymine linked to hypoxanthine or imidazole by a trimethylene chain were studied in aqueous solution. Irradiation (A = 254 nm) of thymine-hypoxanthine pair yielded two internal cycloadducts with azetidine and cyclobutane part structures. Sensitization and quenching experiments suggested that the excited singlet was the reactive state in the photocycloaddition reactions. Only cyclobutanes were isolated from irradiated (A = 290 nm) solutions of thymine-imidazole pairs. Photocycloadditions were reversible upon irradiation at A = 254 nm.

The characteristics of nucleobase transport and metabolism by the perfused sheep choroid plexus.

The uptake of nucleobases was investigated across the basolateral membrane of the sheep choroid plexus perfused in situ. The maximal uptake (U(max)) for hypoxanthine and adenine, was 35.51+/-1.50% and 30.71+/-0.49% and for guanine, thymine and uracil was 12.00+/-0.53%, 13.07+/-0.48% and 12.30+/-0.55%, respectively with a negligible backflux, except for that of thymine (35.11+/-5.37% of the U(ma...

Regulation of de novo purine synthesis in human bone marrow mononuclear cells by hypoxanthine.

In previous studies from this laboratory, human bone marrow hypoxanthine concentrations were found to average 7.1 microM, three times higher than plasma hypoxanthine concentrations measured simultaneously. To assess the significance of this finding, the relationship between hypoxanthine concentration and the rate of purine nucleotide synthesis by the de novo pathway was studied in normal human ...

brush-border membrane vesicles

The characteristics of hypoxanthine transport were examined in purified brush-border membrane vesicles isolated from guineapig kidney. Hypoxanthine uptake in the vesicles was specifically stimulated by both Na+ and an inside-negative potential, resulting in a transient accumulation of intravesicular hypoxanthine. Na+-dependent hypoxanthine influx was saturable (apparent Km 4.4 + 2.1 ,uM, Vmax 1...

Absorption Kinetics of Some Purines, Pyrimidines, and Nucleosides in Taenia Crassiceps Larvae

In vitro kinetic studies revealed that Taenia crassiceps larvae absorb purine bases (adenine and hypoxanthine) and nucleosides (adenosine and uridine) by mediated transport. Pyrimidines (thymine, uracil, and cytosine) are absorbed primarily by diffusion. The presence of hypoxanthine stimulated adenosine uptake, an effect which was enhanced by preloading the worms with hypoxanthine. Furthermore,...

High affinity sodium-dependent nucleobase transport in cultured renal epithelial cells (LLC-PK1).

Two distinct transporters for nucleobases have been characterized in LLC-PK1 cells. The first system accumulates hypoxanthine against a concentration gradient in the presence of sodium. The sodium-dependent uptake of hypoxanthine was saturable at 22 degrees C with a Km value of 0.79 +/- 0.43 microM, a Vmax of 15 +/- 4 pmol/mg protein/60 s, and a Na+:hypoxanthine coupling stoichiometry of 1.27 +...
